These conditions are often associated with the formation and aggregation of neurotoxic misfolded oligomers such as amyloid-β, tau proteins, and α-synucleins. Understanding these harmful oligomers is essential to uncovering the molecular basis of these diseases. To investigate the protein aggregation dynamics and liquid-liquid phase separations, this project will use advanced TEM imaging in Liquid Phase. The research focuses on proteins associated with Alzheimer's and Parkinson's diseases, using a combination of simulations and liquid TEM imaging. Additionally, the project explores the impact of drugs and aggregation inhibitors on protein structures in liquid environments. This interdisciplinary approach combines imaging science, physics, molecular engineering, and computational science and holds great promise for helping to create innovative treatments for neurodegenerative diseases causing dementia.We welcome individuals willing to combine experimental imaging with computational efforts. Proyecto de Generación de Conocimiento 2022 by the Spanish Ministry of Science and Innovation Introduction to the vacant position: The Bioinspired Interactive Materials and Protocellular System Group is looking for a PhD student to develop his/her PhD thesis project on the development of Phagocytic Synthetic Cells, a new type of polymer-based synthetic/hybrid vesicle capable of detecting, engulfing and killing pathogenic bacteria-resistant to antibiotics. The contract will be within the framework of Project PredaSynCell (Generación de Conocimiento). PredaSynCell's objective is to devise and test a new family of synthetic vesicles capable of specific artificial phagocytosis of bacteria. This fellowship is associated with the research project Generación de Conocimiento , funded by the Spanish Ministry of Science and Innovation. Main tasks and responsibilities: Synthesis and molecular characterization of macromolecular amphiphiles. This includes polymers via control radical polymerization techniques as well as Janus dendrimers Synthesis of antimicrobial polymers Study of self-assembly of the aforementioned amphiphiles into vesicles Assembly of vesicles by microfluidic techniques Characterization of the membrane properties by advanced microscopies (optical and force) as well as other techniques. Study of the mechanisms of engulfment and strategies to favor it Study antimicrobial activity Requirements for candidates: Essential: Degree and Master in any field of Chemistry (macromolecular chemistry, synthetic chemistry) Multidisciplinary qualifications (Physics/Math, Physics/Engineering, etc.) and/or knowledge of polymer science (specifically on controlled polymerization and experience with self-assembly will be a plus) as needed for this highly interdisciplinary project. The cell nucleus is a central decision hub in cells, receiving inputs from the cell environment and integrating them to control cell function. In recent years, it has become clear that the nucleus receives not only biochemical signals but also mechanical forces, and that these forces regulate nuclear function. This implies that there are likely multiple regulatory mechanisms deciding when and how forces reach the nucleus, with potential implications in diseases such as cancer or fibrosis, where there are major mechanical alterations to tissues. The candidate will carry out a PhD focused on the process of mechanotransduction, which is the conversion of mechanical forces into downstream biochemical signals (such as for instance transcription). More specifically, the candidate will explore different mechanisms by which cells can regulate whether forces reach or not the nucleus, and for how long. This will be studied in the context of fibroblasts and mesenchymal cells in a pancreatic cancer setting, which is characterized by a dramatic alteration in tissue mechanical properties. This fellowship is associated with the research project Generación de Conocimiento , funded by the Spanish Ministry of Science and Innovation. Main tasks and responsibilities: Work with a model system of fibroblasts due to their robust mechanical responses, and their involvement in pathologies such as cancer or fibrosis, with well-established mechanically-dependent functions (such as matrix remodelling). Establish a toolset for probing ECM-nucleus mechanical coupling, involving mechanical and molecular techniques. Unveil how ECM-nucleus mechanical coupling regulates mechanotransduction. Assess the functional consequences of ECM-nucleus mechanical coupling. Focus on events known to be triggered by mechanical force application to the nucleus. This includes nuclear translocation of transcription factors, activation of calcium signalling, changes in chromatin methylation, and DNA damage.
