The Staff Scientist will prepare grant applications and other scientific communication materials on a wide range of genomic research topics. This role is deeply immersed in the science of NYGC and supports our research team in building and expanding a successful, independently-funded research base. Interested in exploring the role of adipose tissues in mucosal inflammation in a supportive, creative, and inclusive environment? The Grasset lab Our lab’s long-term objective is to uncover novel mechanisms underlying the pathology of Crohn’s disease (CD), a progressive incurable disease characterized by transmural intestinal inflammation leading to complications often requiring bowel resection surgery. The goal is to identify novel therapeutic targets by focusing on two previously disconnected hallmarks of CD: expanded mesenteric adipose tissue wrapping around the intestine – or creeping fat – and anti-microbial antibodies, both correlating with disease complications. Studying these hallmarks will further reveal unexplored relationships between adipocytes, stromal cells, and immune cells driving B cell responses in gut homeostasis and inflammation. Understanding these relationships will impact how we perceive immune responses at other mucosal sites and systemically. Ongoing projects include: the study of the impact of CD-specific microbiota on stromal cell-supported lymphoid structures embedded in mesenteric adipose tissue; specific mechanisms by which stromal cells and adipocytes communicate with B cells; and the subsequent impact on B cell activation and their differentiation to anti-microbial antibody-producing plasma cells. The mission of the Genomics AI Initiative is to address key bottlenecks in genomic interpretation by integrating high-throughput sequencing, single-cell and spatial multiomics, perturbation and functional genomics, and modern machine learning approaches. The Director of Molecular Diagnostics serves as the CLIA Laboratory Director for NYGC's clinical molecular diagnostics laboratory. This role provides clinical and scientific leadership for a CLIA-certified, CAP-accredited laboratory performing next-generation sequencing (NGS)-based genetic and oncology testing. The Director is responsible for the clinical validity, regulatory compliance, and operational excellence of all laboratory testing activities. This includes oversight of assay development and validation, quality management systems, regulatory compliance, personnel competency, and clinical interpretation and reporting of molecular diagnostic tests. The Director ensures compliance with CLIA, New York State Department of Health (NYS CLEP), CAP, and other applicable federal and state regulations, including licensure requirements for laboratories providing testing services across multiple U.S. states. The Zeltser lab is looking for a motivated scientist to join their team as an Assistant Professor of Medical Sciences in Medicine at the Columbia University Medical Center (non-tenure track). The lab studies how the sympathetic nervous system regulates metabolism and how this is impacted by obesity and aging. We utilize a variety of genetically engineered mouse models, virus-based neural tracing and chemogenetic approaches, single nuclear transcriptomics, and in vivo physiological and metabolic measurements. The ultimate goal is to identify neural circuits that regulate distinct aspects of brown fat and function and systemic metabolism. The ideal applicant will be an experienced researcher with expertise in both neuroanatomy and the neurobiology of autonomic regulation of peripheral targets and/or central regulation of metabolic parameters. The procedures for this project require expertise in multiple advanced techniques. The experienced researcher must be able to master state-of-the-art techniques to map and manipulate sympathetic circuits regulating discrete targets in the periphery, including stereotaxic injections, viral tracing, chemogenetics and fiber photometry. In parallel, this project also involves the analysis of physiological phenotypes in vivo, including thermogenesis, glucose uptake and lipolysis. Experience with surgical techniques in mice is essential. The applicant will be expected to lead projects independently and to train and mentor others. The applicant is also expected to make substantive contributions to the formulation and writing of manuscripts and grant applications. Interest in developing new technologies is a plus. The ARPA-H funded project is part of a multi-institutional effort (IndiPHARM) led by Dr Gary Miller from Columbia University, Mailman School of Public Health. The project itself is led by Drs Serge Cremers, and Alex Lyashchenko who will be overseeing the activities of the fellow in the Clinical Pharmacology and Toxicology Laboratory of the Department of Pathology and Cell Biology at Columbia University Irving Medical Center. The work is to lead to several scientific publications and will provide a unique opportunity for clinical pathologists and clinical chemists to gain experience in the development of highly complex mass spectrometry assays and their application in patient care. Research Responsibilities Laboratory-based research efforts will focus on the molecular biology of gene therapy for malignant and non-malignant diseases with the goal of include establishing a platform for tracking clonal expansion and monitoring transgene integration and expression in gene therapy trials, as well as participating in the development of novel strategies for the treatment of globin disorders. Research projects will be conducted under the mentorship of senior investigators at CUIMC and CICET, including Drs. Essential Functions and Accountabilities • Technical and scientific oversight of the laboratory including selection of appropriate test methodology. b. Verification of test procedure analytical performance., • Establish and maintain the quality control program including acceptance criteria throughout the entire process from raw material receipt to reporting of test results., • Resolve technical problems and ensure that remedial actions are taken whenever test systems deviate from established specifications., • Experience in a regulated clinical laboratory environment meeting CLIA/CAP/NYS DOH/ISO 15189/ISO 13485 standards., • Perform hands-on, wet lab experiments to evaluate existing genetic testing assays, leading to assay improvements. Use scientific methods and best practices for experimental protocol design, execution, data analysis, and interpretation. Liaise with R&D team to ensure successful new product introduction by employing rigorous validation protocols and methods., • Analyze and interpret genetic testing output from data analysis pipelines to assess data quality, support the interpretation of sequencing results, and identify trends for optimization. Aid in troubleshooting and root-cause analysis, and devise solutions and approaches for solving technical problems to improve genetic testing performance in an inclusive and collaborative fashion., • Divided between office and clinical laboratory work environment., • Position may at times require the employee to work in areas with bloodborne pathogens or where hazardous materials and/or infectious diseases are present., • At times it will entail prolonged sitting in front of a computer., • In-depth technical knowledge, proficiency and prior experience with molecular biology, genomics, and associated discipline-specific analytical tools, as well as statistical methods is required. Experience with Sanger and Next Generation Sequencing methods and output required., • Hands-on experience with bioinformatics and bioinformatics tools for sequencing data to enable experimental iteration independent of external support will provide an advantage., • Experience in process, method, or assay development and optimization with thorough understanding of verification and validation methods is required.
